The World Gym on Route 22 in Pawling may be the closest and there is also a NYSC located in Carmel. Taking back roads from Holmes to either shouldn't be too bad of a trip.

Unfortunately, nowhere at all. Having known people from Holmes, I can tell you that at least one of them drove all the way out to Wappingers Falls to find one to his liking.
Now, there are others closer than that, but none of them are in Holmes. For example, there's a World Gym out in Pawling, Taconic Fitness in Beekman, and a Gold's Gym in Carmel. For the ladies, there's a Curves in Hopewell Junction.
